Integration of Hydrogen-Deuterium Exchange (HDX) Method with High-Resolution MS
Many harmful immune responses and diseases are believed to be caused by changes in protein conformation. Until now, X-ray crystallography and NMR-based analyses have primarily been used to measure protein conformation. While these analytical methods are useful, they are not applicable to all proteins and often require high-purity, large quantities of samples. Today, by effectively combining the ACQUITY M-Class HDX system with high-resolution mass spectrometry and rigorously controlling the temperature-sensitive aspects of the HDX sample preparation, we can reliably identify changes in protein conformation. This has made it possible to obtain detailed information about protein conformation.
